Sorafenib Formulation
For in vivo experiments: For a concentration of 30 mg/Kg (BW:25g):
1. Heat Cremophor EL at 60°C
2. Dissolve 100 mg of Sorafenib in 2.5mL of a stock solution containing 75% Ethanol and Cremophor EL(1:1)at 60°C (40mg/mL).
3. Vortex at highest speed and put back on 60°C, until Sorafenib is completely dissolved (This process can take up to 15 min).Freeze aliquots and store at -80°C.
4. Dilute the stock solution 1:4 in water and applicate 100 uL orally gavage for each mouse.
5. Note: The sorafenib will flocculate from diluted solution after one or two hours. You need start all over again for each dilute.
For a concentration of 100 mg/Kg (BW:25g):
1. Heat Cremophor EL at 60°C
2. Solve 250 mg of Sorafenib in 10 mL of a stock solution containing 75% Ethanol and Cremophor EL(1:1)at 60°C (25mg/mL).
3. Vortex at highest speed and put back on 60°C, until Sorafenib is completely solved (This process can take up to 20 min). Store at -80°C.
4. Applicate 100 uL orally gavage for each mouse.
